| Subcontractor | Department of Health and Human Services | IN THIS CONTRACT, THE BOYD LAB WILL CARRY OUT THE FOLLOWING ACTIVITIES:
PROJECT 1: CREATE ANTIBODY GENE SEQUENCE LIBRARIES FROM A MICHIGAN HUMAN COHORT AND, SUBJECT TO AVAILABILITY, A
HONG KONG HUMAN SUBJECT COHORT, FROM PERIPHERAL BLOOD MONONUCLEAR CELLS OF SAMPLES TAKEN DURING
SURVEILLANCE FOR INFLUENZA INFECTION, OR PRIOR TO AND FOLLOWING INFLUENZA VACCINATION. THESE LIBRARIES WILL BE
SEQUENCED USING THE ILLUMINA MISEQ INSTRUMENT. DATA WILL BE ANALYZED TO EVALUATE B CELL CLONAL POPULATIONS
AND CANDIDATE VACCINE-RESPONSIVE B CELLS.
PROJECT 2: FROM A HUMAN COHORT IN PENNSYLVANIA, CREATE ANTIBODY GENE SEQUENCE LIBRARIES FROM PERIPHERAL
BLOOD MONONUCLEAR CELLS OF SAMPLES TAKEN DURING SURVEILLANCE FOR INFLUENZA INFECTION, OR PRIOR TO AND FOLLOWING
INFLUENZA VACCINATION. THESE LIBRARIES WILL BE SEQUENCED USING THE ILLUMINA MISEQ INSTRUMENT. DATA WILL BE
ANALYZED TO EVALUATE B CELL CLONAL POPULATIONS AND CANDIDATE VACCINE-RESPONSIVE B CELLS. IN ADDITION, INFLUENZASPECIFIC
B CELLS WILL BE ISOLATED BY FLOW CYTOMETRIC SORTING FOLLOWING INFLUENZA VACCINATION, AND SINGLE-CELL PCR
WILL BE CARRIED OUT TO AMPLIFY ANTIBODY HEAVY AND LIGHT CHAINS. WE WILL THEN SEQUENCE THOSE PCR AMPLICONS AND
CLONE THEM INTO EXPRESSION VECTORS FOR GENERATION OF MONOCLONAL ANTIBODIES. MONOCLONAL ANTIBODIES WILL BE
SHARED WITH OTHER PROJECT INVESTIGATORS FOR CHARACTERIZATION FOR BINDING TO INFLUENZA VACCINE COMPONENTS IN
ELISA ASSAYS AND WITH HEMAGGLUTININ INHIBITION ASSAYS. WE WILL ANALYZE THE ANTIBODY GENE SEQUENCE DATA AND
CORRELATE IT WITH THE BINDING AND OTHER PROTEIN ASSAY DATA FROM THE MONOCLONAL ANTIBODIES GENERATED. IN ADDITION
WE WILL SEQUENCE ANTIBODY GENE LIBRARIES GENERATED FROM FERRETS IN THIS PROJECT, AND ATTEMPT TO CLONE THE HEAVY
AND LIGHT CHAIN GENE SEQUENCES FROM FERRET CELLS THAT BIND INFLUENZA ANTIGENS, TO EXPRESS THE ANTIBODIES AS
PROTEIN FOR FURTHER CHARACTERIZATION BY OTHER PROJECT INVESTIGATORS. WE WILL ANALYZE THE FERRET ANTIBODY GENE
SEQUENCE DATA BY COMPARING THEM TO THE HUMAN ANTIBODY DATA GENERATED IN PROJECTS 1 AND 2.
ALL DATA GENERATED IN THE PROJECT WILL BE SHARED WITH THE OTHER PROJECT AND CORE INVESTIGATORS FOR JOINT ANALYSIS
AND CORRELATION OTHER DATA INCLUDING CLINICAL AND VIRAL ANALYSES. 504429 Parent award context Parent award agency: Department of Health and Human Services · National Institutes of Health Parent award NAICS: 541715 Parent award performance state: PA Parent award description: AWARD OF THE BASE TO UPENN FOR CEIRR CONTRACT 75N93021C00015.
