IN THIS PROJECT, WE BUILD ON PAST FDA FUNDED WORK THAT HAS LED TO THE DEVELOPMENT OF NEW ORGAN-ON-A-CHIP (ORGAN CHIP) MICROFLUIDIC CULTURE DEVICES AS PRECLINICAL TOOLS FOR EVALUATION OF EFFICACY AND SAFETY OF RADIATION MEDICAL COUNTER MEASURES (MCMS), INCLUDING THOSE FOR ACUTE RADIATION SYNDROME.THE MAJOR TASKS ARE TO: 1) TEST THE EFFECTS OF RADIATION MCMS IN THE HUMAN BONE MARROW CHIP RADIOTOXICITY MODEL, 2) EVALUATE THE ROLE OF GUT MICROBIOME IN HUMAN RADIATION INJURY, 3) RECAPITULATE RADIATION-INDUCED PNEUMONITIS IN HUMAN LUNG ALVEOLUS CHIP, AND 4) LEVERAGE THESE IN VITRO MODELS TO DISCOVER NEW RADIATION MCMS. ALL ORGAN CHIPS (BONE MARROW, INTESTINE, AND LUNG) WILL BE EXPOSED TO RADIATION AT BOSTON CHILDREN'S HOSPITAL (BCH), AND EXPERIMENTAL HANDLING OF CHIPS IMMEDIATELY PRIOR AND AFTER EXPOSURE WILL BE CARRIED OUT IN OUR BCH LABORATORY TO ENSURE WE ANALYZE IMMEDIATE AND RAPID RESPONSES AS WELL AS LONGER-TERM BIOLOGICAL EFFECTS AFTER EXPOSURE. THE RADIATION-PNEUMONITIS MODEL IN HUMAN LUNG ALVEOLUS CHIP WILL ALSO BE DEVELOPED, OPTIMIZED, AND STUDIED IN THE BCH LABORATORY WHERE CLINICAL EXPERTS IN LUNG INJURY ARE LOCATED.
