FORGE THERAPEUTICS IS DEVELOPING A PORTFOLIO OF NOVEL, DIRECT-ACTING, SMALL MOLECULE INHIBITORS OF ESSENTIAL METALLOENZYMES FOUND IN HIGH PRIORITY GRAM-NEGATIVE BACTERIA. FORGE€„¢S PLATFORM COMPRISES A DEEP KNOWLEDGE OF METALLOENZYMES, BIOINORGANIC AND MEDICINAL CHEMISTRY KNOW-HOW, AND A FOCUSED FRAGMENT LIBRARY OF PROPRIETARY METAL-BINDING PHARMACOPHORES (MBPS) THAT PROVIDE SELECTIVE AND DIVERSE STARTING POINTS FOR NOVEL INHIBITORS. OUR STRATEGY IS TO FOCUS ON ESSENTIAL METALLOENZYMES FOUND BROADLY ACROSS GRAM-NEGATIVE BACTERIA BUT NOT IN HUMANS. IN BRIEF, OUR LEAD TARGET, LPXC, IS A ZINC METALLOENZYME REQUIRED FOR SYNTHESIS OF THE OUTER MEMBRANE OF GRAM-NEGATIVE BACTERIA. PREVIOUS EFFORTS ON INHIBITING LPXC RELIED ON A HYDROXAMIC ACID (HA) MOIETY TO ANCHOR THE INHIBITOR TO THE CATALYTIC ZINC. ALTHOUGH POTENT IN VITRO, HA-BASED CHEMISTRY SUFFERS IN VIVO FROM GLUCURONIDATION, HYDROLYSIS, AND POOR PK. TO DATE, THERE ARE NO APPROVED ANTIBIOTICS TARGETING LPXC. OUR APPROACH IS TO IDENTIFY NOVEL NON-HA MBPS TO SERVE AS BETTER STARTING POINTS FOR INHIBITOR DEVELOPMENT. FORGE€„¢S GOAL FOR THE FG-LPXC LUNG PROGRAM IS TO DISCOVER AND DEVELOP A NOVEL LPXC INHIBITOR FOR THE TREATMENT OF LUNG INFECTIONS CAUSED BY GRAM-NEGATIVE BACTERIA WITH A FOCUS ON P. AERUGINOSA.
