STATEMENT OF WORK PI: MAƩHEW FRIEMAN UNIVERSITY OF MARYLAND, BALƟMORE PENN CEIRR OPƟON 18.L SEPTEMBER 11, 2024 IN THESE STUDIES, WE WILL DEVELOP AND USE A NOVEL PHAGE DISPLAY CORONAVIRUS SPIKE LIBRARY, SPIKE-SCAN, TO DETERMINE HOW VACCINE PLATFORM, FORMULATION OR COMBINATION SCHEME AƯECTS EPITOPE PRESENTATION. WE HYPOTHESIZE THAT THE DEVELOPMENT OF THIS DATA WILL AID IN OUR FUTURE VACCINE DEVELOPMENT TO DETERMINE HOW EACH OF THESE PARAMETERS AƯECTS EPITOPE SPREAD AND ULTIMATELY FUTURE PROOFING OF VACCINES. THIS NEW “SPIKE-SCAN” PLATFORM WILL BE USED TO ASSESS ANTIBODIES PRODUCED ACROSS MULTIPLE VACCINE PLATFORMS INCLUDING VARIANT SPIKE PROTEINS, NOVEL ADJUVANTS AND HUMAN MIX AND MATCH VACCINE STUDIES CONTROLLED CLINICAL TRIALS TO DETERMINE HOW VACCINE PLATFORM AND FORMULATION AƯECTS BREADTH OF COVERAGE. THE SPIKE-SCAN RESULTS COMBINED WITH TRADITIONAL ELISA AND NEUTRALIZATION STUDIES WILL BE USED TO INFORM FUTURE VACCINE DEVELOPMENT TO PRODUCE WIDER BREADTH AND QUANTIFIABLY HIGHER ANTIBODY RESPONSES. OUR GOAL IS TO DETERMINE AN OPTIMIZED COMBINATION OF ANTIGEN, ADJUVANT AND PLATFORM FOR FUTURE VACCINE DEVELOPMENT FOR CORONAVIRUSES. IN THIS PROJECT WE WILL FIRST PRODUCE A SPIKE DIRECTED CORONAVIRUS “SPIKE-SCAN” LIBRARY THAT INCLUDES ANIMAL AND HUMAN CORONAVIRUS SPIKE PROTEINS INCLUDING CURRENT SARS-COV-2 VARIANT SPIKE SEQUENCES. WE HAVE IDENTIFIED A BROAD SPECTRUM OF SPIKE PROTEINS ACROSS CORONAVIRUS LINEAGES THAT WILL ALLOW FOR DEEP PROFILING OF SERA FROM VACCINE STUDIES. UPON SUCCESSFUL CREATION OF THE SPIKE-SCAN LIBRARY, WE WILL ANALYZE SERUM FROM THE FOLLOWING SUBSETS: 1. NOVAVAX VARIANT SPIKE VACCINES, INCLUDING MULTIPLE SARS-COV-2 VARIANT SPIKES 2. HEXAPRO-WU1 SPIKE WITH VARIOUS ADJUVANTS TO EVALUATE CHANGES IN BREADTH OF COVERAGE 3. IMPRINTING MODELS TO ASSESS IMPACT OF PREVIOUS EXPOSURE 4. MIX AND MATCH VACCINE PLATFORMS (MRNA, ADENOVIRUS AND PROTEIN) FROM HUMAN TRIAL PATIENTS THIS WORK CAN ALSO BE APPLIED IN THE FUTURE TOWARD OTHER VACCINES INCLUDING INFLUENZA AND OTHER RESPIRATORY PATHOGENS THAT UNDERGO MUTATION AND SELECTION IN OUTBREAK SCENARIOS.
