DR. TIROUVANZIAM AND HIS LABORATORY AT EMORY UNIVERSITY COMMITS TO PROVIDING SUPPORT FOR THE FDA GRANT DIRECTED BY DR. SHUICHI TAKAYAMA (THE GEORGIA INSTITUTE OF TECHNOLOGY AND EMORY UNIVERISTY, ATLANTA, GEORGIA, USA), ENTITLED: €œIN VITRO ASSESSMENT OF CHEMICAL MIXTURE- INDUCED AIRWAY INFLAMMAAON IN HEALTHY AND DISEASED LUNGS€Ͽ½. THIS PROJECT WILL USE A HIGH-THROUGHPUT IN VITRO AIR-BLOOD BARRIER ARRAY BASED BARRIER BREAKDOWN AND NEUTROPHIL TRANSMIGRATION/ACTIVATION ASSAY TO PREDICT HUMAN ADVERSE REACTIONS TO INHALED SUBSTANCES. DOSE RESPONSE CURVES AND BENCHMARK DOSES (BMD) WILL BE DETERMINED FOR A RANGE OF WELL-KNOWN AND UNKNOWN SUBSTANCES INCLUDING ELECTRONIC NICOTINE DELIVERY SYSTEMS (ENDS) CONSTITUENTS AND MIXTURES. INITIAL YEARS WILL FOCUS ON HIGH-THROUGHPUT E-CIGARETTE VAPOR EXTRACTS (EVE) ANALYSIS, LATER YEARS WILL ALSO ANALYZE E-CIGARETTE AEROSOL DIRECTLY IN A LOWER-THROUGHPUT FORMAT. THE ORGAN-LEVEL FUNCTIONAL READOUTS (TEER & TRANSMIGRATION) RELATE CLOSELY TO CLINICAL/PRE-CLINICAL BRONCHOALVEOLAR LAVAGE (BAL) MEASURES, ARE CONVENIENT IN CONSISTING OF JUST A FEW KEY NUMBERS, YET ARE INTEGRATIVE OF MULTIPLE MOLECULAR AND CELLULAR PATHWAYS AND THUS HIGHLY PHYSIOLOGICALLY-RELEVANT. THIS PROJECT WILL ANALYZE THE EFFECT OF BIOLOGICALLY VARIABILITY WITH REGARDS TO THE SEX OF THE PRIMARY HUMAN NEUTROPHILS AND EPITHELIAL CELLS. THE PROJECT WILL ALSO ANALYZE THE IMPACT VAPING PLUS OTHER LUNG INSULTS CAN HAVE WHEN COMBINED TOGETHER BY TESTING EVES IN COMBINATION WITH DEFINED INFLAMMATORY COCKTAILS REFERRED TO AS SYNTHETIC AIRWAY FLUID SUPERNATANT (SASN) (EARLY YEARS) OR COPD ASN (LATER YEARS). SUPPORT PROVIDED BY THE TIROUVANZIAM LAB AT ERMORY WILL BE IN THE FORM OF: (I) SUPPORT FOR THE DEVELOPMENT OF THE PRIMARY MODEL; (II) PERFORMANCE OF PHENOTYPIC AND FUNCTIONAL ASSAYS; (III) SUPPORT FOR DATA ANALYSIS AND INTEGRATION; AND (IV) REPORTING OF RESULTS AND CONTRIBUTION TO SCIENTIFIC ABSTRACTS AND MANUSCRIPTS. THE TIROUVANZIAM LAB IS PARTICULARLY WELL EQUIPPED FOR SUCH STUDIES AND HAS UNIQUE EXPERTISE IN THE IMPLEMENTATION OF IN VITRO ASSAYS RELATED TO AIRWAY NEUTROPHLIC INFLAMMATION, AS EVIDENCED BY PRIOR PUBLICATIONS RELEVANT TO THIS TOPIC. TO ENSURE PROPER IMPLEMENTATION OF THE EXTENSIVE WORK INCLUDED IN THIS STATEMENT, DR. TIROUVANZIAM WILL COMMIT 10% OF HIS EFFORT, 10% OF EFFORT FOR A POSTDOCTORAL BIOINFORMATICIAN AND 20% FOR A RESEARCH SPECIALIST SUPERVISOR IN EACH OF YRS 1-3 OF THIS PROJECT.

THIS STUDY WILL UTILIZE SYSTEMS IMMUNOLOGY APPROACHES TO INVESTIGATE THE IMMUNE SIGNATURES GENERATED BY OUR SARNA YFV VACCINE AND COMPARE THESE TO THE GOLD STANDARD YFV-17D LIVE-ATTENUATED VACCINE (YF-VAX). IMMUNE SIGNATURES WILL BE OBTAINED BY TRANSCRIPTOMIC ANALYSIS FOLLOWING PRIME AND BOOST IMMUNIZATIONS AND WILL CAPTURE BOTH EARLY INNATE AND ADAPTIVE IMMUNE SIGNATURES. IN ADDITION, WE WILL INVESTIGATE THE ANTIBODY REPERTOIRE GENERATED BY OUR VACCINE PLATFORM. WE WILL CORRELATE THESE SIGNATURES TO RESPONSE MEASURES OF CELLULAR AND HUMORAL IMMUNITY. CELLULAR IMMUNITY WILL FOCUS ON MARKERS OF MEMORY DEPOSITION FOR B CELLS (E.G., CD27) AND T CELLS AS A PRELIMINARY INDICATOR OF RESPONSE DURABILITY AND LONGEVITY.
