| Subcontractor | Department of Health and Human Services | IN THIS PROJECT, WE WILL STRUCTURALLY AND BIOPHYSICALLY CHARACTERIZE NEUTRALIZING ANTIBODIES AND
IMMUNE SERA DERIVED FROM NATURAL INFECTION BY INFLUENZA VIRUS AND FROM VACCINATION. THE WILSON
LAB WILL EXPRESS AND PURIFY ANTIBODIES, ANTIGENS AND IMMUNOGENS FROM MAMMALIAN CELL LINES,
INCLUDING 293S, 293T AND 293F CELLS, DROSOPHILA CELLS, OR E. COLI FOR STRUCTURAL AND BIOPHYSICAL
STUDIES. WE WILL THEN STRUCTURALLY AND BIOPHYSICALLY ANALYZE THESE NEUTRALIZING ANTIBODIES WITH
INFLUENZA ANTIGENS [HEMAGGLUTININ (HA), AND NEURAMINIDASE (NA)], AS WELL AS IDENTIFIED RESISTANCE MUTANTS, TO DETERMINE ANTIBODY AFFINITIES AND MODE OF BINDING, STABILITY, STOICHIOMETRY, EPITOPES AND 3D STRUCTURES. WE WILL USE X-RAY CRYSTALLOGRAPHY TO DETERMINE THE HIGH-RESOLUTION STRUCTURES OF ANTIBODIES AND ANTIBODY-ANTIGEN COMPLEXES. OTHER BIOPHYSICAL METHODS INCLUDING BIO-LAYER INTERFEROMETRY (BLI), ISOTHERMAL TITRATION CALORIMETRY ITC), DIFFERENTIAL SCANNING CALORIMETRY (DSC) AND SURFACE PLASMON RESONANCE (SPR), WILL BE USED TO DETERMINE BINDING AFFINITIES AND PROTEIN/IMMUNOGEN STABILITY. WE WILL DESIGN, MODIFY AND CHARACTERIZE IMMUNOGENS AS VACCINES CANDIDATES USING STRUCTURE-ASSISTED VACCINE DESIGN METHODS THAT WE HAVE PREVIOUSLY SUCCESSFULLY UTILIZED FOR HIV, INFLUENZA VIRUS AND HEPATITIS C VIRUS. THE WARD LAB WILL UTILIZE THEIR RECENTLY DEVELOPED NOVEL EM IMAGING METHOD OF POLYCLONAL EPITOPE MAPPING (EMPEM) TO ANALYZE POLYCLONAL ANTIBODY RESPONSES IN SERA TO IDENTIFY PATTERNS OF EPITOPE UTILIZATION AND FREQUENCY IN HA AND NA IN HUMAN LONGITUDINAL COHORTS. THE HA OR NA PROTEIN PROBES IN THESE ANALYSES WILL BE PRODUCED IN THE WILSON LAB AND SERA PROVIDED BY OUR COLLEAGUES IN THIS CEIRR PROJECT. 504429 Parent award context Parent award agency: Department of Health and Human Services · National Institutes of Health Parent award NAICS: 541715 Parent award performance state: PA Parent award description: AWARD OF THE BASE TO UPENN FOR CEIRR CONTRACT 75N93021C00015. | Subcontractor | Department of Health and Human Services | KYLIE KONRATH WILL VISIT THE WARD LAB FOR THE PURPOSE OF LEARNING OUR EMPEM PIPELINE FOR MAPPING THE EPITOPES OF POLYCLONAL ANTIBODIES ON VIRAL ANTIGENS. SHE WILL LEARN HOW TO PREP SERUM, PURIFY IGG, DIGEST INTO FAB, AND MAKE IMMUNE COMPLEXES. SHE WILL ALSO LEARN HOW TO PREPARE SAMPLES FOR IMAGING IN THE ELECTRON MICROSCOPE AND COLLECT DATA. THESE DATA WILL THEN BE PROCESSED USING 2D AND 3D CLASSIFICATION TO GENERATE 3D MAPS OF THE IMMUNE COMPLEXES WHICH CAN BE USED TO INTERPRET THE IMMUNOGENIC SURFACE OF INFLUENZA ANTIGENS THAT ARE TARGETED AFTER VACCINATION WITH IMMUNOGENS DESIGNED IN THE KULP LAB. 0258-A460-4609 (DR.
